Inhibit the γ-secretase complex, indirectly inhibiting the Notch pathway with DAPT. This notch pathway inhibitor promotes the differentiation of human pluripotent stem cells into nociceptors (Chambers et al. Nature Biotechnol, 2012) and pancreatic cells (D'Amour et al. Nature Biotechnol, 2006). It is also used to promote the differentiation of mouse embryonic stem cells to neurons (Crawford and Roelink. Dev Dyn, 2007) and retinal pigment epithelium (Osakada et al. Nat Protoc, 2009). DAPT also reduces colony-forming efficiency of mouse neural stem cells (Androutsellis-Theotokis et al. Nature, 2006) and reduces mammosphere-forming efficiency of breast cancer cell lines and ductal carcinoma in situ cells (Farnie et al. J Natl Cancer, 2007; Harrison et al. Cancer Res, 2010).
